A more t h o r o u g h i n v e s t i g a t i o n o f t h e b i n d i n g o f t h i s and o t h e r phenot h i a z i n e drugs t o phospholipid liposomes i s r e p o r t e d here. The e q u i l i b r i u m b i n d i n g o f c h l o r p r o m a z i n e , t h i o r i d a z i n e , p r o c h l o r p e r a z i n e and p e r p h e n a z i n e t o u n s o n i c a t e d liposornes formed from s o y - b e a n p h o s p h a t i d y l c h o l i n e was s t u d i e d a t 25+1 OC i n subdued a r t i f i c i a l lighting. The b i n d i n g was d e t e r m i n e d u n d e r t h e s e c o n d i t i o n s by e q u i l i b r i u m d i a l y s i s u s i n g V i s k i n g t u b i n g . B i n d i n g d a t a w e r e a n a l y s e d b y b o t h t h e l i n e a r method of S c a t c h a r d ( 1 9 4 9 ) and t h e n o n - l i n e a r method o f Thompson & K l o t z (1971 1. L i n e a r S c a t c h a r d p l o t s i n d i c a t e d simple b i n d i n g t o i d e n t i c a l s i t e s f o r a l l t h e d r u g s , and g a v e v a l u e s of k , t h e i n t r i n s i c a s s o c i a t i o n c o n s t a n t , and l / n , t h e number of l i p i d m o l e c u l e s bound p e r m o l e c u l e of d r u g ( T a b l e 1 ) These b i n d i n g p a r a m e t e r s f o r chlorpromazine a r e s i m i l a r t o t h o s e o b t a i n e d b y Kovaleva & o t h e r s ( 1 9 7 5 ) , u s i n g a l i n e a r method o f a n a l y s i s . However, t h e more i n f o r m a t i v e Thompson & K l o t z a n a l y s i s i n d i c a t e d a complex t y p e o f b i n d i n g f o r a l l t h e drugs, except prochlorperazine. A c c o r d i n g t o t h e Thompson & K l o t z p l o t s , t h i s complex b e h a v i o u r c o u l d be a t t r i b u t e d t o a n i n t e r a c t i o n b e t w e e n b i n d i n g s i t e s , b e i n g m o s t l y of t h e ' o b s t r u c t i v e ' t y p e , a l t h o u g h t h e r e was some e v i d e n c e of ' c o - o p e r a t i v e ' b i n d i n g f o r low c o n c e n t r a t i o n s of chlorpromazine. Binding complexity i s a l s o r e f l e c t e d i n t h e d i s c r e p a n c y between t h e b i n d i n g p a r a m e t e r s d e t e r mined b y t h e two methods ( T a b l e I ) , and t h e f a i l u r e t o o b t a i n v a l u e s o f t h e s e f o r c h l o r p r o m a z i n e by t h e Thompson & K l o t z method. S i m i l a r complex b e h a v i o u r h a s b e e n o b s e r v e d i n t h e b i n d i n g of t h e s e d r u g s t o liposomes p r e p a r e d from b r a i n l i p i d s .
