Such coinsiderations ledc us to attemil)t recove ry of a component frori-i remienrtumr exl)osedl to a periodontal pocket that wouLld react with seruim antibocly fromn the same individual (EVIERHART et al. Abstract 55 (Special Issue) No. 645, 1976). Fturther obseraztions in this stuidy ar e herewith reported. Twenty-five teeth were extracted frorn patients (age rarnging from} 30 to 65 years) who ) had sevei periodontal disease. Derntal extractions under local anesthesia ancd blood sampling were done at the same tirme. After extraction. cementLumii above the still-attached fibers was r-emoved by root planing. Some dentin obviously was mixedl with each of the specimen sai-nples. Cementuim was washed in 5 nil of saline and removecl by centriftugation at 1.000 X g at 4 C; this supernatant was Si. Ceinenturm was then placed in-glycine-HCl buffer (1)H 2.5) with I M NaCl. rmixed for 20 minut',,s and centrifogated as above. The supernatant (S2) was then homogeniied in a glass tissuie grinder containing horate-saline (pH 8.2) and centrifugated as described, with a resuiltanrt stipernatant, S:. All stipernatants were dialyzed three timnes against dlistilled water at 4 C and lyophilized. Samples were reconstituited in borate-saline. As controls, cementurm from fouir totally impacrted teeth was subjected to procedures identical to those outlined. Countercurrenit immnuinoelect rophoresis was uLsed to show the antibody-antigen reacrtion. Agarose (0.8) was dissolved in barbital buffer (pH 8.2) and 10 Inls )laced on a 75 X 50 mm glass slicle whirh was eleetrophoresed uising barbital buffer (pI- 8.2) and 140 V and 20MA per slide for 20 Tninuties. Pairs of wells werc cut in the a-ar 6imiii apart. The serum was added to the well nearest the positixve pole anId the cementtiiii sarrinle added to the other well. Ouir resuilts showed S., to be the sample with the antigenie component. The reactions of two S-, sarrples froiii cernentum exposed to a perio-

donitail pocket with homologouLs serLuln are shown in 1 and 2 of the figure: 3 shows the reaction of S. with heterologous sera froin pcrsons suffering with periodontal disease. These sera were negative with the S. Of impacted cementum. Our data, therefore, demonstrated that a comj)onent from pocket-exposed cementum will react with homologouis serum and with heterologous sera from persons suffering with periodontal disease. The reactive component removed fiom pocket-exposed cementum appeared to be absoibed to the surface, since the low p1)r high salt buffer uised to remove the component wotuld not be exp'cted to remove chemically couLpled material. In suimmary, an antigen can be removed from cementum exposed to a periodontal pocket. Since only adults were used in this stuLdy. we were uinable to obtain and inluctde in this sttudv erupted teeth without some form of gingival or periodontal disease. However, an an-tigen was not present on cementuns fromn totally impacted teeth, possibly becauise this ceementurn was never in contact with the oral cenironment. Our findings imply that the antigen recovered was not a normiial component of cemnentum. Thus, the immune responses seen in pcriodontal disease may not be autoimmtine, but rather rnay reprcsent a response to a foreign antigen. Obviously, such a factor may be significant in initiatinig the imTmlLune response an-d the tissue damage seen in inflam,imatory periodontal disease.
